The unique morphological manifestations called “ Mycorrhizae” represents one of the nature’s most widespread phenomenon of symbiosis. Global Warming is one of the challenges that we are facing in today’s era. The effect of global warming is not only limited to human beings but its effect can also be seen on plants. In this report we have used data mining techniques to show that heat stress is an important stress affecting the agricultural production. At the molecular level post translational modifications have been shown to be influenced in plants under heat stress. We have studied Palmitoylation, Sumoylation, S- Nitrosylation and Nitration in the plant gene sequences associated with heat stress and not associated with heat stress. We show that sumoylation is an important property which is possibly influenced in large number of proteins associated with heat stress in a wide variety of plants. This property can be used for identifying novel sequences associated with heat stress in plants.

The medicinal plant Sutherlandia Frutescens (SF) is commonly used in South Africa, and its popularity stems from its use as a cancer treatment. It is also endorsed by the South African Ministry of Health as a supplementary treatment for HIV and AIDS. Maize is the staple crop of South Africa, and can be frequently contaminated by the mycotoxin Fumonisin B1 (FB1). Although little is known about its role in human disease, FB1 has been epidemiologically linked to oesophageal cancer in South Africa. Both SF and FB1 have been shown to promote apoptosis, yet the effect(s) of consuming both in combination is currently unknown. The aim of this study was to determine whether SF and FB1 had either synergistic or antagonising effects in combination, by investigating immune cell toxicity in Jurkat cells. Apoptotic parameters such as caspase activation, mitochondrial depolarisation, phosphatidylserine (PS) externalisation and ATP quantification were analysed. Oxidative stress parameters were also investigated. The results suggest that SF induces apoptosis, characteristic of its nature as an anti-cancer treatment, and FB1 induces oxidative stress, characteristic of its carcinogenic properties.

The so-called creep strength enhanced ferritic 9-12% Cr steels have been identified as the most promising class of materials for some of the key components in ultra-supercritical fossil-fired power plants, including the main steam pipes, headers and superheater tubings. These steels are less costly, and they have a lower coefficient of thermal expansion and a higher thermal conductivity when compared with austenitic stainless steels, making them less susceptible to degradation through thermal fatigue. However, experience has shown that the weldments in these steels are particularly prone to premature creep failure, due to a localised form of cracking in the heat-affected zone, which is referred to as Type IV cracking. The work presented in this thesis is concerned with the effects of residual stresses and constraint on Type IV cracking. It was found that the highest as-welded tensile stresses resided near the outer boundary of the HAZ, and towards the weld root region and these were not fully relieved by the applied PWHT. In both conditions substantial tensile direct and hydrostatic stresses existed across the HAZ, including the fine-grained and intercritically-annealed regions.

The current work aimed at achieving a better understanding about the effect of stress factors on plant gene expression. In the first part a search for genes up-regulated in response to drought/osmotic stress was carried out by using a drought-tolerant wheat (Triticum aestivum L.) cultivar. Through a subtraction molecular approach differentially expressed genes were selected that could be further used in strategies for improving drought tolerance in crops. The second part of the work was focused on the regulation of alfalfa (Medicago sativa L.) B-type cyclin-dependent kinase (Medsa;CDKB2;1). A fragment from the kinase upstream gene region was cloned and shown to be sufficient to assure a cell cycle-dependent and G2/M-phase-specific gene expression, similarly to the endogenous CDKB2;1 kinase. Living organisms are continually exposed to oxidants. During oxidative stress, reactions of oxidants with biological macromolecules can result in both direct damage and the formation of long-lived, reactive, hydroperoxides. The series of studies undertaken in this work focused on the early cellular events following generation of reactive singlet oxygen (1O2) and identified that long-lived protein hydroperoxides, a major product of protein oxidation, can play a significant role in cellular damage by selectively targeting both low-molecular mass and protein thiols. These findings are invaluable in providing an insight of the early molecular events of oxidative stress and a cornerstone for future research in developing a better strategy to prevent oxidative damage.

Oxidative stress has been implicated in the progression of several diseases ranging from mild inflammation to several inflammatory acute immune diseases to grave disease process like cancer.The steady state levels, or extent of accumulation of oxidation products in tissues and plasma or the changes in the antioxidant status are commonly used as measures of increased oxidative stress. In fact, the measurement of lipid peroxides, conjugated dienes, and markers of nitrative DNA damage are considered to be the cornerstone of the assessment of oxidative damage in vivo. Oral Lichen Planus (OLP) is an inflammatory disease of unknown etiology while Oral Lichenoid Reaction (OLR) is a condition mimicking OLP. As these conditions are exposed to oxidative stress, they could release reactive oxygen species (ROS) which are implicated in the pathogenesis of a plethora of inflammatory conditions to lethal diseases. We evaluated and compared the levels of a series of oxidative stress markers in patients with OLP and OLR with that of normal controls and tried to identify the role of these oxidative stress markers in these conditions.

Although the United States Ready Reserve has not received much attention in the past, they are getting it now, given its more active interactions on foreign and domestic public policies. Hence, this research has sought to reflect upon the relationship between stress and supervisory styles in the reserve centers. This study reviewed and critiqued relevant empirical literature of the past two decades concerned with stress in the workplace. It also visited some historical publications on supervision and leadership styles affecting performances. In all, the results showed that the organizational culture is more related to stress than the supervisory styles in a naval reserve center.

The thyroid hormones play a pivotal role in the generation of oxidative stress on exposure to stressor. It was further observed that vitamin E treatment eliminated the stress induced oxidative stress while the raised levels of T3 and T4 remained unaffected.The present investigations provided enough evidence to show that the supra spinal response to stress basically elevates the thyroid hormone levels, which onward causes the oxidative stress. Such conclusion further suggests the use of antithyroid agent to prevent the stress induced oxidative stress and subsequent tissue degeneration. However, it is cautioned that the levels of thyroid hormones must be first assessed in stressed exposed subjects before any antithyroid agent is used to prevent the oxidative stress as the hypothyroidism after the inappropriate dose of antithyroid agent is likely to impair the stress- sustainace ability.

Presence of heavy metals in the soil and water is an inevitable evil in modern day due to industrialization. Heavy metal stress is one of the major abiotic stresses that cause environmental pollution in recent decades. These metals unlike other organic pollutants are not degraded and converted into harmless compounds via biological processes. Hence, they can persist for a long time in the environment and enter into food chain. The effects of aluminium and zinc on growth and Biochemical components were investigated in 20 days old seedlings of Pennisetum americanum (L). Leek and Phaseolus aureus in Hoagland’s solution spiked with various concentrations of aluminium and zinc. Total chlorophyll content, glucose and protein declined progressively with increasing the concentration of heavy metals. A significant increase of proline, amino acids and other enzymes were detected in primary leaves after exposure to heavy metals. The strongest effect on these chemicals was found in plants exposed to Aluminium.

AM fungi are helpful to enhance growth and productivity in BT cotton under salt and water stress conditions. Three genera and 33 species were isolated from rhizosphere soil of BT cotton. The genus Glomus was dominant with 19 species, nine species of Scutellospora and five species of Acaulospora were recorded from surveyed sites. Mycorrhizal plants produced more number of leaves than control plants,they were taller than control plants,the roots were also longer in mycorrhizal plants than control plants.The cotton bolls, picked bolls, weight of bolls and weight of seed cotton per boll of mycorrhizal BT cotton was much better than control plants. Plants treated with mycorrhizae protected plants from water and salt stress up to certain limit as compared to control plants. The AM fungi limited the increase in number of Verticillium dahliae propagules.

The current study investigated the relationship between workload and occupational stress among managers in manufacturing industries. Data were collected from 40 managers who were selected from Unilever, Goldstar Sugars, Astra Paints and Innscor. Survey questionnaires were used to collect data and these were administered to all selected managers with the aim of investigating the relationship between workload and occupational stress. Results of the study showed that workload is significantly related to occupational stress among managers. It also emanated from the study that workload contribute significantly to overall feelings of work-related stress and that work-related stress manifests itself psychologically, behaviourally, emotionally and physically.

In saline soils, banana suffered from salt injury with the external symptoms. The caroteniod content was decreased in all the treated plants except in control. In all the NaCl treated plants the chlorophyll pigments was significantly reduced. In abscissic acid(ABA), treated plants chlorophyll content was significantly increased. Membrane Stability Index(MSI) was decreased when the salt stress period increased. In case of Cholorophyll Stability Index(CSI) and Membrane Stability Index(MSI), the butylated hydroxyl toluene(BHT) increased over the other chemicals treated in NaCl stress, where as in acetyl salicylic acid (ASA) increased MSI only. The ASA capable of inducing more ascorbic acid production in more NaCl stressed plants. Peroxidase and superoxide dismutase activities were decreased significantly over the period of stress increases. In BHT treated salt stressed plants observed significantly higher nitrate redcutase activity. The persent investigation showed that the abscissic acid(ABA), acetyl salicylic acid (ASA) and butylated hydroxy toluene (BHT) have the potential to reverse the effect of salinity in Grandnain banana plants.

Drought stress is one of the most common environmental abiotic stress that affects the overall growth and development of plants. Comparative analysis was carried out between the transgenic plants (Gossypium hirsutum) inherited a gene related to small heat shock protein gene (sHSP) family (GHSP-26) and wild type (non-transgenic plants under different morphological, physiological and biochemical level. The GHSP-26 gene was isolated from the Gossypium arboreum which is one of the most tolerates species of cotton to different stress condition. In this study we also identified and analysed drought responsive genes in both genotypes with the help of Real time PCR. To carry out this task, RNA was extracted from the promising lines under drought stress from which cDNA was synthesized. This cDNA was used in Real Time PCR for the Relative quantification of our genes of interest. Here we also provide the detail protocols of the parameters required for the screening of the drought tolerance genotypes.

Improving tolerance to heat stress is a major challenge in many crops, especially C3 crops, given the threat of recent global warming. While significant achievements have been made on understanding the molecular and physiological mechanisms to short–term exposure to high temperature, the physiological mechanisms of summer tolerance in fields, which are characterized by prolonged exposure to high temperature stress, are not still well understood. In this study, six experiments were conducted to clarify the physiological mechanisms of tolerance to high temperature stress by comparing between genotypes, populations and different species including C3 and C4 photosynthetic types. Overall, although the complexity of heat tolerance was revealed by the involvement of various mechanisms such as suppression of ROS, scavenging of toxic ROS, and leaf functions, this study definitively showed that the oxidative stress resulting from the excessive production of ROS is the main cause of damages under heat stress.

The U.S financial crises of 2008 highlighted the role of a depressed financial system in causing an economic downturn. This pioneer empirical study develops a financial stress index for the Pakistan economy during 2000-2013. We use key variables that capture unique aspect of stress in the financial system. Principal component analysis technique is used to aggregate the components to a single index. Empirical analysis reveals that banking sector and stock market volatility are the leading contributors to financial stress in Pakistan. The developed financial stress index is then used to trace high episodes of financial stress. Our index was successful in identifying three main financial crises that attacked the financial system including global financial crisis of 2008. Afterwards, the impact of financial stress on real economic activity is examined using impulse response function and Granger causality analysis. We find the expected results as high values of financial stress index are associated with decline in macroeconomic indicators. These findings will provide useful information to readers about the financial and economic dynamics of a developing economy like Pakistan.

Lead is an environmental toxic metal, which is capable of causing many chronic diseases. Important sources of lead include pipes, lead containing paints, gasoline containing lead, and the different kinds of canned foods. In the present study lead toxicity and the oxidative stress caused by lead intoxication has been studied. Oxidative stress is a very major effect of lead. The mechanism of lead-induced oxidative stress involves an imbalance between generation and removal of ROS (reactive oxygen species) in tissues and cellular components causing damage to membranes, DNA and proteins. The blood samples from lead exposed and control males were collected and sera was isolated from them. Different biochemical tests were performed on serum by the use of biochemical kits. The results indicated an increase in glucose concentration in lead exposed subjects.
